Isolated/virtual environments are not 100% fail safe, hence I use a dedicated test system on which my VM/Sandboxie Is running.

check downloaded items on virustotal , etc

Sandboxie for sure. There have been a lot of alternatives. But, this is plain and simple.
For advanced virtualization, I just fire the Virtualbox and throw anything highly suspicious.

I agree with your response. But, I've had no issues testing few viruses on Qubes OS.

I agree with your response. But, I've had no issues testing  few viruses on Qubes OS.

Exactly what Issues are you experiencing?

Install a Windows OS on the VM, and you will not come across any Issues.

My preferred method is to simply build it from source after verifying the tarball signature. sometimes that's not possible, so I resort to just verifying the signature and using sandboxd.

Same, If the file is from unknown source, I will always scan it and run in sandbox.

Malware Is known to circumvent a Sandbox environment, so as an added layer of protection, run It In a VM, preferably on a dedicated Host physical machine.
